Missions are a gameplay element in Resident Evil: Revelations that players can complete to unlock extra items. These missions can be accessed from the main menu. Upon completing a mission, a brief announcement is displayed during gameplay. However, the unlockable item is not received until the player manually 'unlocks' it from the mission menu by pressing the A button over the mission. Completed missions will show an exclamation mark, and once unlocked, they will move to the end of the list and be darkened.

Missions requiring a certain task to be done a specific number of times carry over across Campaign Episodes and Raid Mode Stages. Progress for these missions is displayed when the mission is highlighted. Unlocked campaign weapons or custom parts will automatically appear in the workbench menu.

Both Campaign mode and Raid mode have their own sets of missions, but they both contribute to the total number of missions completed, which is displayed. Cleared missions and certain Raid Mode missions can be deleted from the mission screen. This may be necessary for the Raid Mode page as new missions cannot be received when all 25 mission slots are full.
