- 1A Vacation In The Wilds: Defeat all five main bosses in Inkwell Isle IV (The Moonshine Mob, Esther Winchester, Glumstone The Giant, Mortimer Freeze, The Howling Aces). This is story-related and cannot be missed.
- 2Ranger: Achieve an A-Rank or higher on all bosses in Inkwell Isle IV. Requirements include defeating the boss in under two minutes on Regular difficulty, having at least 3HP remaining, landing three parries, and six special attacks. Secret bosses count, and using Djimmi The Great's wishes for more HP voids progress.
- 3Alive And Kicking: Defeat any boss with Ms. Chalice. Equip the Astral Cookie charm and defeat any main boss from Inkwell Isle I to IV, including DLC bosses and original favorites.
- 4Decadent: Defeat 10 bosses with Ms. Chalice. This applies only to main bosses. Equip a new DLC weapon and defeat all bosses on Inkwell Isle I and II on Simple or Regular difficulty.

- 8A King's Admiration: Defeat the King's Gauntlet. This challenge requires defeating all five mini-bosses sequentially without failing. Failing restarts the entire gauntlet. Ms. Chalice is recommended.
- 9Compliments To The Chef: Complete your quest on Inkwell Isle IV by defeating all five main bosses and collecting ingredients for the Wondertart. This unlocks the secret antagonist, Chef Saltbaker, for a final boss fight. Crackshot, Ms. Chalice, and determination are recommended.
- 10Cooked To Perfection: Achieve an S-Rank on a stage in Inkwell Isle IV. S-Ranks are only available on Expert difficulty (unlocked after finishing the main story and dealing with The Devil). Criteria are the same as A-Ranks. The Moonshine Mob is suggested if you don't mind its minions.
- 11The High Hat: Defeat a boss on Inkwell Isle IV without killing any of its minions. Esther Winchester in "High Noon Hoopla" is a good candidate, as the only killable henchman is the mini-cow.
- 12Hearty: Have 9HP at one time. Equip the Twin Heart charm and use Djimmi The Great's wish for an additional 3HP. Alternatively, try strategies outside King Dice's Casino or combine the Heart Ring charm with Djimmi for 9HP after six successful parries. Ms. Chalice is highly beneficial.
- 13A Horrible Night To Have A Curse: Survive the nightmare (Secret). Obtain the Broken Relic charm from Porkrind's Emporium and use it for the riddle near the southeast side of Isle IV.
- 14Paladin: Obtain great power (Secret). Completing "A Horrible Night To Have A Curse" unlocks the Cursed Relic (1HP, random weapon). Beating seven or more bosses with it on Regular or Expert transforms it into the Divine Relic (3HP, added abilities).
