Resident Evil 3: Nemesis features Mikhail, Nicholai, Murphy Seeker, Tyrell Patrick, Barry Burton, Chris Redfield, Leon Kennedy, Claire Redfield, and Sherry Birkin.

Mikhail Victor: A Captain in the U.B.C.S. mercenary forces, Mikhail was formerly at odds with the Russian military, leading to a death sentence. Umbrella negotiated his release and brought him into their ranks. He was deployed to Raccoon City with Carlos and, after heavy losses and injuries, retreated into a streetcar with survivors.

Nicholai Ginovaef: A cold, calculating Sergeant in the U.B.C.S., Nicholai previously served in the Soviet Army and was part of GRU's Spetsnaz. By September 1998, he was within Umbrella's paramilitary hierarchy. In Raccoon City, he appears focused on escape, but his true agenda is sinister.

Murphy Seeker: An infected U.B.C.S. sniper found near death in the Umbrella Sales Office. Depending on the player's path, Murphy is killed by either Carlos or Nicholai before he can become a zombie.

Tyrell Patrick: Another U.B.C.S. mercenary who knows more about Umbrella's operations than most. Depending on player choices, he either dies opening a booby-trapped safe in the Hospital basement or kills himself with a grenade.

Barry Burton: A S.T.A.R.S. Alpha Team comrade of Jill and a survivor of the Spencer mansion incident. He felt guilt over being blackmailed by Albert Wesker, who threatened Barry's family. After Wesker's threat was revealed as a bluff, Barry knocked out Wesker and helped other S.T.A.R.S. members escape. He moved his family to Canada for protection but rushed back to Raccoon City upon hearing of the outbreak, hoping to rescue survivors.

Chris Redfield: A former U.S. Air Force pilot and S.T.A.R.S. Alpha Team survivor, Chris is known for his temper and marksmanship. He bonded with Jill during their time in the Spencer mansion. After escaping and attempting to expose Umbrella, Chris suspended himself from the R.P.D. to investigate Umbrella's European headquarters.

Leon Kennedy: A rookie cop who encountered a zombie outbreak on his first day with the R.P.D. He met Claire Redfield, who was searching for her brother Chris. Together, they navigated the police station and sewers, reaching Umbrella's NEST facility before its destruction. They also rescued Sherry Birkin.

Claire Redfield: Chris Redfield's sister, a college student who traveled to Raccoon City after losing contact with him. She encountered zombies before meeting Leon. They sought refuge in the R.P.D., unaware it was overrun. Claire befriended Sherry Birkin, who was infected with the G-Virus by her father. Claire secured a vaccine to prevent Sherry's mutation. After escaping Raccoon City, Leon agreed to watch over Sherry, allowing Claire to continue her search for Chris.

Sherry Birkin: The young daughter of Umbrella researchers Dr. William Birkin and Dr. Annette Birkin. Following her parents' deaths and her escape with Leon and Claire, Sherry was taken into government custody after Leon became a U.S. government agent in exchange for her guaranteed protection.
