- Rune #1: Purchase from Piero for 500 coins. He only has one available.
- 2Rune #2: Found on a small piece of land along the shoreline after gaining powers from The Outsider and visiting The Hound Pits for the first time. Use Blink to reach it.
- 3Runes #3 & #4: After investigating sewer noises post-High Overseer Campbell, find one Rune behind a grating underwater at the end of a tunnel, and the other inside the desk near the Weepers.
- 4Rune #5: Awarded by Pendleton if you duel Lord Shaw during Lady Boyle’s Last Party.
- 5Rune #6: Received via a letter on the table next to Corvo’s bed if you dispatch Lady Boyle non-lethally during Lady Boyle’s Last Party.

- 7High Overseer Campbell:
- Rune #1: In the first main area, follow the path to the right towards a balcony, then head downstairs and out the door next to Granny Rags. The Rune is at an Outsider Shrine at the end of the path.
- 8Rune #2: Reward for completing Granny Rags' first side mission.
- 9Rune #3: Reward for completing Granny Rags' second side mission.
- 10Rune #4: Located inside a trailer-like building on the left side of the area past the second Wall of Light.
- 11Rune #5: Found in a secret room within the High Overseer's Office. Access the room by pushing the eye on a statue at the base of the east set of stairs leading to the second floor. The Rune is inside a glass case that must be broken.
- 12Rune #6: Located in the Meeting Room in the Office of the High Overseer, hanging above the fireplace.
- 13Rune #7: Found inside the Workshop building in the Back Yard area, on a table. Enter by breaking the glass on the roof and dropping inside.
- 14House of Pleasure:
- Rune #1: Inside Galvani’s Offices in the second area of the Distillery District, within the secret lab. Pull a specific book on the bookshelf to open it.
- 15Rune #2: Located inside the Art Dealer’s safe in the Art Dealer’s Apartment. Obtain the combination by electrocuting the Art Dealer in the Golden Cat Bathhouse during Slackjaw’s second side mission. The combination changes each playthrough.
- 16Rune #3: In the Golden Cat area, on the second floor of a building just to the right of the main entrance stairs, on a table.
- 17Rune #4: On the second floor of the bathhouse. From the top of the stairs, go right through the double doors. The Rune is hanging above a table on the wall.
- 18Rune #5: Located on the top floor of The Golden Cat, in the room with the Courtesan, next to where Emily is held.
- 19The Royal Physician:
- Rune #1: Past the initial guards and the area with the cart and Warehouse Key, on a street with more guards. On the right is a gateway leading to stairs and a chain. Climb the chain and enter the room to find the Rune on a table.
- 20Rune #2: Inside a safe in Pratchett’s house in Drawbridge Way. The combination is 473, found by observing three paintings in a specific order. The safe is on the top floor.
- 21Rune #3: From Pratchett's house, go upstairs and onto the roof. Blink onto the balcony overlooking the waters.
