Dead or Alive 6 includes many returning fighters from previous installments, alongside new characters. Notably, this installment marks the first time characters from outside the Dead or Alive franchise have been included. Team Ninja collaborated with SEGA to bring reworked versions of Virtua Fighter characters into the game, and these can be unlocked relatively early.

The base game features a total of 24 characters. Additionally, there is potential for downloadable characters (DLC (downloadable content — extra missions or items released after launch)) to be released post-launch.
